PENGARUH KOMBINASI UKURAN EXPLANT, PEMANASAN DAN PENGGUNAAN RIBAVIRIN PADA PERTUMBUHAN JARINGAN MERISTEMATIK BAWANG MERAH
Abstract
An experiment was conducted in Tissue Culture Laboratory of IVEGRI in Lembang. It aimed to observe effect of heat treatment, antiviral Ribavirin and explant size on growth of merismatic tissue of shallot. Medium used was MS added with 30g sucrose, 2mg IAA, 2mg kinetin, 0.1mg GA3 and 6g agar per Liter with pH 5.7. Treatments consisted of three factors, namely explant size (of meristem and of shoot tip); heat treatment (36ºC for one month and without heat treatment); and antiviral Ribavirin addition (5 mg L-1 and no- Ribavirin). Results showed that heat treatment decreased contamination percentage in culture. It was observed that there was no interaction between the three factors examined, although visually explant size and heat treatment affected growth. Explant growth was very affected by explant size, heat treatment and antiviral Ribavirin.
